چکیده

Here we tried to develop a standard flow cytometry protocol for measuring the physiological state of the Gram +‟ve bacterium Bacillus cereus, and attempted to understand more of the resulting staining phenomena using cell sorting. Specifically the use of DiOC6(3) and the proprietary RedoxSensor Green TM (an indicator of bacterial reductase activity) both counter stained with propidium iodide. The data presented here has identified at least 4 distinguishable physiological states based on the two staining protocols alone and dependent on the position in the growth cycle single cells gave rise to varying numbers of colonies when sorted individually onto nutrient agar plates. Further that these growing colonies derived from single cell sorts had widely different lag phases. This highlights further the unexpectedly complex population dynamics of bacterial cultures and lays to rest the microbiological dogma that all bacterial cells in a culture behave in exactly the same way since they are clearly members of a physiologically heterogenous and dynamic population.

A vector for promoter trapping in Bacillus cereus.

We constructed a promoter-trap plasmid, pAD123, for Bacillus cereus. This plasmid contains a promoterless gene that encodes a mutant version of the green fluorescent protein, GFPmut3a, that is optimized for fluorescence-activated cell sorting [Cormack, B.P., Valdivia, R.H., Falkow, S., 1996. FACS-optimized mutants of the green fluorescent protein (GFP). Gene 173, 33-38.]. The plasmid replicates...

Purification of Immune Cell Populations from Freshly Isolated Murine Tumors and Organs by Consecutive Magnetic Cell Sorting and Multi-parameter Flow Cytometry-Based Sorting.

It is well established that tumors evolve together with nonmalignant cells, such as fibroblasts, endothelial cells, and immune cells. These cells constantly entangle and interact with each other creating the tumor microenvironment. Immune cells can exert both tumor-promoting and tumor-protective functions. Detailed phenotypic and functional characterization of intra-tumoral immune cell subsets ...

Analysis of bacterial function by multi-colour fluorescence flow cytometry and single cell sorting.

With the increased awareness of the problems associated with the growth dependent analysis of bacterial populations, direct optical detection methods such as flow cytometry have enjoyed increased popularity over the last few years. Among the analyses discussed here are: (1) Bacterial discrimination from other particles on the basis of nucleic acid staining, using sample disaggregation to provid...

P-95: Flow Cytometry Analysis of Bovine Semen:A Qualitative Study

Background: Although AI practices have been introduced little over 60 years, the success rate remains relatively low. This might be due to the exclusive selection of semen based on motility analysis. Recent advancement in sperm sexing using flow cytometry with an increased throughput from next generation cell sorters, made use of this technology in studding sperm qualitative aspects other than ...

Optical Trapping of Individual Human Immunodeficiency Viruses in Culture Fluid Reveals Heterogeneity with Single-Molecule Resolution

Optical tweezers use the momentum of photons to trap and manipulate microscopic objects, contact-free, in three dimensions. Although this technique has been widely used in biology and nanotechnology to study molecular motors, biopolymers and nanostructures, its application to study viruses has been very limited, largely due to their small size. Here, using optical tweezers that can simultaneous...
